Investigation of convergent and divergent genetic influences underlying schizophrenia and alcohol use disorder

Background
Alcohol use disorder (AUD) and schizophrenia (SCZ) frequently co-occur, and large-scale genome-wide association studies (GWAS) have identified significant genetic correlations between these disorders.
MethodsWe used the largest published GWAS for AUD (total cases = 77 822) and SCZ (total cases = 46 827) to identify genetic variants that influence both disorders (with either the same or opposite direction of effect) and those that are disorder specific.
ResultsWe identified 55 independent genome-wide significant single nucleotide polymorphisms with the same direction of effect on AUD and SCZ, 8 with robust effects in opposite directions, and 98 with disorder-specific effects. We also found evidence for 12 genes whose pleiotropic associations with AUD and SCZ are consistent with mediation via gene expression in the prefrontal cortex. The genetic covariance between AUD and SCZ was concentrated in genomic regions functional in brain tissues (p = 0.001).
ConclusionsOur findings provide further evidence that SCZ shares meaningful genetic overlap with AUD.

Competition from weeds is one of the major biophysical constraints to rice (Oryza spp.) production in sub-Saharan Africa. Smallholder rice farmers require efficient, affordable and labour-saving weed management technologies. Mechanical weeders have shown to fit this profile. Several mechanical weeder types exist but little is known about locally specific differences in performance and farmer preference between these types. Three to six different weeder types were evaluated at 10 different sites across seven countries – i.e., Benin, Burkina Faso, Côte d'Ivoire, Ghana, Nigeria, Rwanda and Togo. A total of 310 farmers (173 male, 137 female) tested the weeders, scored them for their preference, and compared them with their own weed management practices. In a follow-up study, 186 farmers from Benin and Nigeria received the ring hoe, which was the most preferred in these two countries, to use it during the entire crop growing season. Farmers were surveyed on their experiences. The probability of the ring hoe having the highest score among the tested weeders was 71%. The probability of farmers’ preference of the ring hoe over their usual practices – i.e., herbicide, traditional hoe and hand weeding – was 52, 95 and 91%, respectively. The preference of this weeder was not related to gender, years of experience with rice cultivation, rice field size, weed infestation level, water status or soil texture. In the follow-up study, 80% of farmers who used the ring hoe indicated that weeding time was reduced by at least 31%. Of the farmers testing the ring hoe in the follow-up study, 35% used it also for other crops such as vegetables, maize, sorghum, cassava and millet. These results suggest that the ring hoe offers a gender-neutral solution for reducing labour for weeding in rice as well as other crops and that it is compatible with a wide range of environments. The implications of our findings and challenges for out-scaling of mechanical weeders are discussed.

The failure of glyphosate to control all weeds throughout the entire growing season has sometimes prompted growers to use herbicides other than glyphosate on glyphosate-resistant soybean. Field studies were conducted in 1999 and 2000 to investigate potential crop injury by several herbicides in glyphosate-resistant soybean and to determine the relationships between soybean maturity group, planting date, and herbicide treatment on soybean injury, leaf area index (LAI), and yield. Glyphosate-resistant soybean generally recovered from early-season herbicide injury and LAI reductions; however, some treatments reduced yield. Yield reductions were more common in double-crop soybean than in full-season soybean. In full-season soybean, most yield reductions occurred in the early-maturing ‘RT-386’ cultivar. These yield reductions may be attributed to reduced developmental periods associated with early-maturing cultivars and double-crop soybean that often lead to reduced vegetative growth and limited LAI. Reductions in LAI by some herbicide treatments were not necessarily indicative of yield loss. Further yield reductions associated with herbicide applications occurred, although soybean sometimes produced leaf area exceeding the critical LAI level of 3.5 to 4.0, which is the minimum LAI needed for soybean to achieve maximum yield. Therefore, LAI response to herbicide treatments does not always accurately indicate the response of glyphosate-resistant soybean yield to herbicides.

We present the results of two 2.3 μm near-infrared (NIR) radial velocity (RV) surveys to detect exoplanets around 36 nearby and young M dwarfs. We use the CSHELL spectrograph (R ~ 46,000) at the NASA InfraRed Telescope Facility (IRTF), combined with an isotopic methane absorption gas cell for common optical path relative wavelength calibration. We have developed a sophisticated RV forward modeling code that accounts for fringing and other instrumental artifacts present in the spectra. With a spectral grasp of only 5 nm, we are able to reach long-term radial velocity dispersions of ~20–30 m s−1 on our survey targets.

Objective.
To evaluate the impact of an institutional hand hygiene accountability program on healthcare personnel hand hygiene adherence.
Design.Time-series design with correlation analysis.
Setting.Tertiary care academic medical center, including outpatient clinics and procedural areas.
Participants.Medical center healthcare personnel.
Methods.A comprehensive hand hygiene initiative was implemented in 2 major phases starting in July 2009. Key facets of the initiative included extensive project planning, leadership buy-in and goal setting, financial incentives linked to performance, and use of a system-wide shared accountability model. Adherence was measured by designated hand hygiene observers. Adherence rates were compared between baseline and implementation phases, and monthly hand hygiene adherence rates were correlated with monthly rates of device-associated infection.
Results.A total of 109,988 observations were completed during the study period, with a sustained increase in hand hygiene adherence throughout each implementation phase (P<.0001) as well as from one phase to the next (P < .0001), such that adherence greater than 85% has been achieved since January 2011. Medical center departments were able to reclaim some rebate dollars allocated through a self-insurance trust, but during the study period, departments did not achieve full reimbursement. Hand hygiene adherence rates were inversely correlated with device-associated standardized infection ratios (R2 = 0.70).
Conclusions.Implementation of this multifaceted, observational hand hygiene program was associated with sustained improvement in hand hygiene adherence. The principles of this program could be applied to other medical centers pursuing improved hand hygiene adherence among healthcare personnel.
Effect of selenium doping on structural and optical properties of SnS:Se thin films by electron beam evaporation method

SnS nanoparticle has been synthesized initially using SnCl2 · 2H2O and Na2S · XH2O, in the presence of TEA by precipitation method and XRD and FTIR techniques have been used for characterization of the sample. Powder X-ray diffraction studies revealed the particle size to be 48 nm and the pattern represents polycrystalline herzenbergite orthorhombic crystal structure of SnS. The FTIR result also confirmed the SnS at 2354 cm−1. Secondly SnS:Se thin films have been deposited on glass substrates by electron beam evaporation technique and the films were annealed at 100 °C and 200 °C for 1 h. The unannealed films are amorphous in nature and the annealed film shows that a sharp crystalline peak is due to SnS. Also a peak is shown at 2θ = 14.39°, which is due to characteristic peak of SnSe2, established by their XRD patterns. The band gap energy (Eg) was determined from transmission spectra and an optical band gap of Eg varies from 1.6 eV to 1.79 eV.

Reported zinc, but not copper, intakes influence whole-body bone density, mineral content and T score responses to zinc and copper supplementation in healthy postmenopausal women

A supplementation trial starting with 224 postmenopausal women provided with adequate vitamin D and Ca was conducted to determine whether increased Cu and Zn intakes would reduce the risk for bone loss. Healthy women aged 51–80 years were recruited for a double-blind, placebo-controlled study. Women with similar femoral neck T scores and BMI were randomly assigned to two groups of 112 each that were supplemented daily for 2 years with 600 mg Ca plus maize starch placebo or 600 mg Ca plus 2 mg Cu and 12 mg Zn. Whole-body bone mineral contents, densities and T scores were determined biannually by dual-energy X-ray absorptiometry, and 5 d food diaries were obtained annually. Repeated-measures ANCOVA showed that bone mineral contents, densities and T scores decreased from baseline values to year 2. A priori contrasts between baseline and year 2 indicated that the greatest decreases occurred with Cu and Zn supplementation. Based on 5 d food diaries, the negative effect was caused by Zn and mainly occurred with Zn intakes ≥ 8·0 mg/d. With Zn intakes < 8·0 mg/d, Zn supplementation apparently prevented a significant decrease in whole-body bone densities and T scores. Food diaries also indicated that Mg intakes < 237 mg/d, Cu intakes < 0·9 mg/d and Zn intakes < 8·0 mg/d are associated with poorer bone health. The findings indicate that Zn supplementation may be beneficial to bone health in postmenopausal women with usual Zn intakes < 8·0 mg/d but not in women consuming adequate amounts of Zn.

In this study, compact Charpy impact testing was used to investigate the effect of processing history and dendrite morphology of bulk metallic glass matrix composites (BMGMCs) on impact toughness. Composite samples were fabricated via suction casting and semisolid forging, and the results were compared with crystalline alloys in the same geometry. A strong dependence on processing was observed, with samples exhibiting up to a 30-fold increase in impact toughness depending on processing and microstructure. Provided that attention is paid to processing techniques, BMGMCs are shown to have properties that equal or surpass some conventionally used crystalline alloys. These properties invite further exploration of these materials in structural applications.
